چکیده

Stealth nanoparticles made of polymer micelles are ideal drug carriers for targeted drug delivery, but their stability is critical for intravenous applications. We found that core-surfacecrosslinked nanoparticles (SCNp) made from amphiphilic brush copolymers had greatly enhanced stability. The surface of the SCNps can be easily tailored by using different chains. The amphiphilic brush copolymers consisted of hydrophobic poly(caprolactone) (PCL) and hydrophilic poly(ethylene glycol) (PEG) or poly(2-(dimethylaminoethyl methacrylate) (PDMA) chains were used to fabricate SCNps. Positively charged PCL/PDMA SCNps could be internalized more efficiently by ovarian cancer cells, leading to an enhanced anticancer drug efficacy.

Blood-stable, tumor-adaptable disulfide bonded mPEG-(Cys)4-PDLLA micelles for chemotherapy.

Although targeted delivery mediated by ligand modified or tumor microenvironment sensitive nanocarriers has been extensively pursued for cancer chemotherapy, the efficiency is still limited by premature drug release after systemic administration. Herein we report a highly blood-stable, tumor-adaptable drug carrier made of disulfide (DS) bonded mPEG-(Cys)(4)-PDLLA micelles. Intravenously injecte...

Bioreducible cross-linked core polymer micelles enhance in vitro activity of methotrexate in breast cancer cells.

Polymer micelles have emerged as promising carriers for controlled release applications, however, several limitations of micelle-based drug delivery have also been reported. To address these issues, we have synthesized a functional biodegradable and cytocompatible block copolymer based on methoxypoly(ethyleneglycol)-b-poly(ε-caprolactone-co-α-azido-ε-caprolactone) (mPEG-b-poly(εCL-co-αN3εCL)) a...

Hydrolysable core crosslinked particle for receptor-mediated pH-sensitive anticancer drug delivery.

Biodegradable micelle systems with both extracellular stabilities and specific targeting properties are highly desirable for anti-cancer drug delivery. Here, we report a biodegradable and crosslinkable poly(propylene fumarate)-co-poly(lactide-co-glycolide)-co-poly(ethylene glycol) (PPF-PLGA-PEG) copolymer conjugated with folate (FA) molecules for receptor-mediated delivery of doxorubicin. Micel...

Surface charge switchable and core cross-linked polyurethane micelles as a reduction-triggered drug delivery system for cancer therapy

The extracellular stability versus intracellular drug release and the long blood stream circulation versus enhanced cell uptake are two dilemmas for micellar drug delivery systems. To resolve the above two problems, a novel core cross-linked polyurethane micelles with redox sensitive and pH-responsive surface charge switchable properties were prepared and investigated as anti-cancer drug carrie...

Spray-dried casein-based micelles as a vehicle for solubilization and controlled delivery of flutamide: formulation, characterization, and in vivo pharmacokinetics.

Novel casein (CAS)-based micelles loaded with the poorly soluble anti-cancer drug, flutamide (FLT), were successfully developed in a powdered form via spray-drying technique. Genipin (GNP) was used to crosslink CAS micelles as demonstrated by color variation of the micelles. Drug solubilization was enhanced by incorporation within the hydrophobic micellar core which was confirmed by solubility ...
